Cholinesterase and phosphatase activities in adults and infective-stage larvae of levamisole-resistant and levamisole-susceptible isolates of Haemonchus contortus

Se detectaron actividades de colinesterasa (ChE) y fosfatasa ácida (AP), pero no de fosfatasa alcalina, en fracciones citosólicas y unidas a membrana de larvas adultas y en etapa infecciosa de Haemonchus contortus resistente y susceptible a levamisol. A diferencia de otros nematodos gastrointestinales, la actividad de ChE fue mayor en L3 que en los adultos y, en ambos casos, estuvo asociada principalmente con las membranas. La actividad de ChE fue inhibida por Triton X-100 y solo se detectó en fracciones unidas a membrana cuando se eliminó el detergente. Se observaron diferencias entre L3 resistentes y susceptibles en la respuesta a los inhibidores (fracción citosólica) y en el contenido enzimático (fracción unida a membrana). La actividad fosfatasa se detectó a pH ácido en todas las fracciones, siendo mayor en el adulto que en el estadio L3. En el primero, la mayor parte de la enzima se localizó en las fracciones unidas a la membrana, mientras que en el segundo se localizó principalmente en las fracciones citosólicas. Esta diferencia podría correlacionarse con la actividad en el intestino. En los ensayos de inhibición, se observó una diferencia entre las fracciones citosólicas de adultos resistentes y susceptibles en su respuesta a 1 mmol/L de ácido tartárico. star_border

Abstract
Cholinesterase (ChE) and acid phosphatase (AP) activities, but not alkaline phosphatase activities, were detected in cytosolic and membrane-bound fractions of adult and infective-stage larvae of levamisole-resistant and levamisole-susceptible Haemonchus contortus. In contrast to other gastrointestinal nematodes, the ChE activity was higher in L-3 than in adults and, in both cases, was mainly associated with membranes. ChE activity was inhibited by Triton X-100 and was only detected in membrane-bound fractions when the detergent was removed. Differences between resistant and susceptible L-3 were observed in the response to inhibitors (cytosolic fraction) and in the enzymatic content (membrane-bound fraction). Phosphatase activity was detected at acidic pH in all fractions, being higher in the adult than in the L-3 stage. In the former, most of the enzyme was localized in the membrane-bound fractions, whereas in the latter it was mainly in cytosolic fractions. This difference could be correlated with the activity in the gut. In inhibition assays, a difference between cytosolic fractions from resistant and susceptible adults was observed in their response to 1 mmol/L tartaric acid.
