Abstract
The tumor necrosis factor (TNFa) is a protein that belongs to the family of cytokines secreted by immune system cells. Its function is to promote inflammatory reactions against infectious agents and tumours. When this protein increases locally due to a pathogen, starts an acute inflammation which will stop after the cause disappears. However, there are pathologies in which the TNFa may be in a permanent overexpression, which results in a constant inflammatory reaction that causes damage to the patient. In this work we review the structure of TNFa, its function and its participation in various pathologies involved. We will discuss about the various therapies in terms of pharmacokinetics, efficacy and mechanism of action. Also we classify treatments based on their structure, which may be antibodies against TNFa or soluble receptors. Finally we will discuss the immunogenicity of treatments and study ways to reduce the appearance of anti-drug antibodies.
